| Further information: | Pichia pastoris GTS115 was grown in minimal SDC medium, consisting of 0.67% bacto-yeast nitrogen base without amino acids, 2% dextrose and 0.2% cas amino acids. The host strain Escherichia coli K12 MC1061 or DH5α can be used for electroporation to amplify the library. The transformation mixture can then be streaked on selective solid medium (LB-Lennox + 100 µg/ml ampicillin) and cultivated at 37°C to obtain the different library clones. |
